994 Effect of dupilumab on the skin mycobiome in atopic dermatitis: A pilot study

Atopic dermatitis (AD) is considered associated with microbial dysbiosis. Dupilumab effective in improving AD, along restoring the skin bacterial microbiome, but its effect on mycobiome have not been investigated. In this prospective study, we aim to study of dupilumab fungal microbiome patients AD. Eight adult moderate severe AD were enrolled. All received monotherapy 300 mg every other week, and swabs collected from lesional nonlesional sites at weeks 0, 4, 8, 12, 16. age- sex-matched non-AD volunteers recruited as controls. DNA extraction amplicon sequencing ITS1 region performed. Our results revealed that most had significant improvement clinical scores after 16 weeks. The alpha diversities, measured by Chao1 indices, significantly lower than control samples. Malassezia was abundant genus healthy skin, relative abundance decreased week 0 during treatment (P < 0.05). abundances globosa sympodialis higher controls lesions (no statistical significance), they showed a decreasing trend treatment. Additionally, found positive correlations between severity including EASI (rho = 0.30, P 0.05), IGA 0.33, 0.001) NRS 0.35, P< 0.01) for data highlight patients.
